This position will support projects to discover and characterize new antiviral factors. Lab tasks including CRISPR knock outs, PCR, DNA sequencing including next generation library preps, cell culture experiments, transformations, ligations, bacterial culture, minipreps, transfections, Western blot. Antiviral immune responses studied may include those that inhibit HIV, ZIKA and/or Coronaviruses. Data and samples must be well-organized and precisely tracked. The most critical qualities in a successful candidate will be organization, precision, and initiative.
Under the direction and close supervision of the Principal Investigator or higher-level laboratory personnel in the lab, the applicant will gain practical experience in the following areas:
coordinating and performing support for a research laboratory that focuses on understanding innate immune responses to HIV and other viral infections
performing state of the art CRISPR screening, in some cases in primary cell systems and validation of genes identified in such screens. Other basic molecular biology techniques will be developed as part of this work, including including learning the basics of molecular cloning, PCR, DNA sequencing, RT-PCR, flow cytometry, cell culture and transfection, and data analysis.

Perform CRISPR KO screens and individual KOs
Culturing of cell lines and primary T cells
Prepare libraries for Illumina sequencing and analyze deep sequencing data
Perform RNA seq
Perform Western blots and other methods to measure gene/protein inactivation
Perform molecular biology techniques such as PCR, RT-PCR, and ddPCR
Assist with molecular cloning and tissue culture for a variety of projects, including studies of viral antibodies and the viral reservoir
Maintain laboratory records and compile and verify the accuracy of data on experiments and assays
Present experimental results in both oral and written report
Perform other related duties as assigned
Keep up to date on literature related to research projects
A bachelors degree in Biology, Chemistry, or Biochemistry and bench research experience:
Research Technician I minimum qualifications - Must have scientific knowledge in the area of study and basic laboratory skills. Independent research experience in addition to school lab experience is usually necessary.
Research Technician II minimum qualifications - Intermediate level position requiring 2 years previous experience working in a laboratory. Independent research experience outside of laboratory course work is usually necessary.
Must have sound laboratory skills, including molecular techniques and cell culture
Must demonstrate exquisite organizational skills, laboratory precision, reliability and work initiative required to operate with minimal daily supervision
Willingness to work in a HIV laboratory after suitable training (BSL2+)
Scientific knowledge of virology, immunology and/or RNA sequence analyses
Some experience with large data sets
Previous laboratory experience with experiment design, execution, optimization, troubleshooting, and data analysis
Demonstrated competence in careful and critical experimental design and execution.
Previous research experience in a BSL 2 laboratory, including tissue culture work
